See that "thesecretofmonkeyisland_save" folder?
Put the savegame in there. You should be able to find it in the savegames screen of monkey island then.

no you don't put it in any "thesecretofmonkeyisland_save" folider, you place it in the directory with the game, where the EXE files is
then smash F5 inside the game

 Before you open DOSBox, copy the saved game into the directory with the game in it. Then, and only then, run the game in DOSBox.
